常用针灸穴位。属足阳明胃经，是合穴。《素问·针解篇》说："所谓三里者，下膝三寸也。"国际标准代号为ST36。
定位 正坐屈膝，穴在犊鼻下3寸，胫骨前缘外一横指。
取穴 ①犊鼻下一夫，胫骨前缘外侧一横指。②手心对髌骨(病人的左手对左脚，右手对右脚)，手指朝向下，无名指指端处即是穴。③从胫骨粗隆处沿胫骨前缘向下摸，胫骨凹陷处外一横指即是穴(程莘农取穴法)。④伸足，脚前伸，胫骨上前缘旁肌肉凹陷处即是穴(《灵枢·邪气脏腑病形》："取之三里者，低跗。")。
刺灸法 直刺1～1.5寸；可灸或针上加灸，一般灸壮较多或灸的时间较长。
功用 补气行气，调理脾胃，疏通经络，清理水湿。
主治 胃痛，腹泻，体弱，水肿(本穴被认为是人体的强壮穴，主治范围很广，一切虚证均可用其来治疗)。
