To investigate the effect of irradiation on the quantity and osteogenesis potential of BMMSCs and to explore the response of them in the irradiation stress and its contribution to long-term effects of radiation-induced bone and hematologic injury, a total body irradiation (TBI) murine model was adopted.

The aim of this study was to explore the changes in cellular senescence related indexes of bone marrow mesenchymal stem cells(BMMSCs) after total body irradiation(TBI).
为了探讨小鼠骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMMSCs)在放射损伤后细胞衰老(cellular senescence)的细胞和分子水平相关指标的变化,本研究采用全身照射(total body irradiation,TBI)小鼠模型,观察4 Gy TBI后4周内不同时间点小鼠BMMSCs的形态学和衰老相关β-半乳糖苷酶(senescence-associatedβ-galactosidase,SA-β-gal)的变化,用流式细胞术分析TBI对BMMSCs细胞周期分布的影响,用实时定量RT-PCR检测细胞衰老相关基因p16INK4a、p21Cip1/Waf1、p53和TGF-β1在TBI前后的表达变化。

为观察异种胸腺修饰对大鼠抗豚鼠天然抗体产生的影响 ,将供体豚鼠和受体SD大鼠随机分成对照组 (a组 )、全身照射组 (b组 )、照射 +胸腺注射组 (c组 ) ,a组大鼠腹腔内注射经处理的豚鼠脾淋巴细胞 (5× 10 6个 ) ,7d后流式细胞仪法检测抗豚鼠IgM及IgG类天然抗体水平的变化 ,b、c组在腹腔注射异种抗原前 17d接受全身照射 ,c组大鼠另在全身照射后 3d接受胸腺内豚鼠脾淋巴细胞注射。

The goal of this study was to investigate the mechanism of discordant xenotransplantation rejection by the study of the effect of cobra venom(CCV)and whole body irradiation(WBI)on guinea pig to rat heart transplantation rejection.
供体豚鼠和受体SD大鼠各 32只随机分成空白对照、单用中华眼镜蛇蛇毒CCV (0 4mg/kg )、单用全身照射WBI (5Gy)、CCV与WBI合用 4组进行异位心脏移植 ,结果表明 :(1)存活时间CCV +WBI组 (12 36± 4 5 3h ) >CCV组 (7 0 6± 3 3h ) >WBI组 (0 42± 0 0 7h )和空白对照组 (0 36± 0 16h ) ,差别显著 ;WBI组与空白对照组无明显差别 (P >0 0 5 ) ;(2 )供心停跳后与移植前比较 :空白对照组血清IgG升高、IgM下降 ,而其余 3组IgG和IgM均下降 ;(3)病理检查空白对照组与WBI组呈超急性排斥 (HAR )改变 ;CCV组和CCV +WBI组呈延迟性异种排斥 (DXR )改变。
