性质:指能与特定DNA或RNA互补结合、抑制基因表达的RNA片段。在复制水平上,反义RNA与RNA 引物互补结合抑制复制;当反义RNA与mRNA5′非编码区或翻译起始部位结合时,可使mRNA不能与核糖体结合或阻止核糖体沿mRNA移动;与真核基因初始转录产物各加工位点;如5′端帽结构形成位点、内含子外显子结合位点、多聚A形成位点等结合时,则影响mRNA的加工成熟和从核内向胞浆转移,从而达到专一抑制基因表达。这将成为寻找低毒、高选择性抗肿瘤抗病毒药物的途径。在合适的启动子和终止子之间,反向插入靶基因,可构建表达反义RNA的重组体,在细胞内表达反义RNA而抑制靶mRNA的表达。反义RNA可以通过重组DNA技术和人工合成法得到。天然反义RNA调控系统广泛存在于原该生物中。
