将含有细胞毒抗体的受者血清与供者的淋巴细胞加入补体后一起培养。受者血清中含有对抗供者淋巴细胞抗原HLA的抗体时,则两者结合后激活补体,损害供者淋巴细胞膜或引起细胞溶解。通过显微镜下观察死亡的淋巴细胞数量,可了解供受者之间的组织相容性。一般要求死亡细胞少于15%。若高于15%,移植后可能出现超急性排斥反应。
