[Objective]To investigate the expression of β-catenin and β-catenin/Tcf complex, two key members in the Wnt/β-catenin pathway, in different differentiated nasopharyngeal carcinoma (NPC ), and elucidate the role of Wnt/β-catenin pathway in neoplastic cells differentiation of NPC.
【目的】探讨Wnt/β-catenin通路的枢纽分子β-catenin和β-catenin/Tcf复合体在不同分化程度鼻咽癌中的表达,进而阐明Wnt/β-catenin通路对鼻咽癌分化的作用。

Expression of β-catenin,Survivin and C-myc involved in Wnt/β-catenin signaling pathway were detected by immunohistochemistry.
材料与方法:将SW480细胞经皮下接种Balb/c-nu/nu裸小鼠,构建裸鼠SW480细胞荷瘤动物模型,观察CPP作用后移植瘤体积和重量的变化,HE染色光镜下观察移植瘤组织形态学变化,免疫组织化学方法检测瘤组织内Wnt/β-catenin信号通路中核心成分β-catenin及其下游靶蛋白Survivin和C-myc表达的变化。

Objective: The inhibition effect of periplocin extracted from cortex periplocae (CPP) on proliferation of human colon carcinoma cell line SW480 in vitro and in vivo was studied, and the regulation effect of CPP on Wnt/β-catenin signaling pathway and its downstream target genes was investigated.
目的:观察香加皮杠柳苷(periplocin extracted from cortex periplocae, CPP)对人结肠癌细胞株SW480在体内外增殖的抑制作用,研究其对SW480细胞内Wnt/β-catenin信号转导通路的调控作用和对β-catenin/TCF下游靶基因的影响,以探讨CPP的抗肿瘤作用机制,为CPP作为抗肿瘤药物开发提供理论依据。

This study was aimed to quantitatively detect the levels of beta-catenin and cyclin D1 mRNA in various subgroups of acute myeloid leukemia(AML)and to analyze their potential relationship,so as to provide theoretical basis for exploring the role of Wnt/beta-catenin pathway in the pathogenesis of AML.
结论:在AML患者中存在beta-catenin及cyclin D1的过度表达,且在部分亚型中存在显著相关性;Wnt/beta-catenin通路在AML中被异常激活,很可能是通过该通路下游靶基因cyclin D1的激活,参与白血病细胞的周期紊乱和异常增殖的调节。

性质: 原癌基因编码的蛋白质,参与钙依黏附蛋白(cadehrin)胞内结构域介导的信号转导的重要分子。正常细胞内它的浓度较低。在由它介导信号转导的结果中都发现APC(adenomatous polyposis coli,是一种肿瘤抑制蛋白质),是一个β-连蛋白信号转导的负调节剂。显然β-连蛋白在恶性肿瘤病因研究领域中占有相当重要的地位。一旦APC或β连蛋白发生突变,即可发生信号转导减弱,细胞增殖加快与抑制细胞凋亡等作用,这是至今所了解的正常细胞发生恶变的β-连蛋白作用分子机制。
