Methods:The modified plate culture method was used to establish biofilm model in vitro with which the morphology of PAO1 biofilms were observed by argentation (ARG) after treated with different dosages of fosfomycin and aminoglycosides.
方法:采用改良的组织培养平板法建立铜绿假单胞菌PAO1生物被膜模型,应用银染法快速鉴定法观察经不同剂量的两类药物单用与联合作用处理后的PAO1生物被膜结构,对其形态学进行描述,并采用计算机图像处理系统进行图像分析。

Objective: To study the biology characteristics of PAO1 and PAO-JP2 and to investigate the influence of quorum sensing on pathogenicity as well as drug tolerance to Pseudomonas aeruginosa; Comparing the biofilm morphology, causative agent and transcriptional level of AHLs between PAO1 and PAO-JP2 which were cultured with diffenent dosages of roxithromycin(ROM), Azithromycin.
目的:本课题通过对比铜绿假单胞菌野生株PAO1及密度感知系统(QS)las和rhl基因缺失的双变异株PAO-JP2的生物学特性,探讨QS系统对铜绿假单胞菌致病性、耐药性的影响;并通过对经罗红霉素、阿奇霉素、乙酰螺旋霉素处理后的PAO1、PAO-JP2生物被膜的形态、致病因子以及lasR基因转录水平比较,探讨阿奇霉素等大环内酯类药物对QS系统在铜绿假单胞菌生物被膜致病性、生物被膜的形成以及在转录水平干扰密度感知信号因子AHL的合成的影响。

Objective: Study the biology characteristics as well as the biofilm morphology of PAO1 and PAO-JP2, and identify the sensitivity of PAO1 and PAO-JP2 in planktonic or in bacterial biofilms to different kinds of antibiotics.
目的:本课题通过对铜绿假单胞菌野生株PAO1及密度感知系统(QS)1as和rh1基因缺失的双变异株PAO-JP2的生物学特性、其生物被膜形态学、不同种抗菌药物对其浮游菌和生物被膜菌杀菌活性的比较研究,探讨QS系统对铜绿假单胞菌致病性、耐药性的影响;并通过对经阿奇霉素处理后的PAO1、PAO-JP2生物被膜的形态学比较,探讨阿奇霉素对QS系统在生物被膜形成方面的影响。
