性质: 指双螺旋DNA溶液中50%的分子受热变性时的温度。符号Tm。这里所指热变性主要是双链核酸解链或成为任意螺旋构象。在不同温度下双链DNA变性程度不同,对紫外吸收值(A260)亦不同。测定各种不同温度下的紫外光吸收值,取其最高吸收值与最低吸收值之间的中点值的温度即为Tm值。在一定的离子强度和酸碱度的溶液中,Tm值与DNA中鸟嘌呤和胞嘧啶两种碱基的含量呈线性关系。所以通过Tm值可以了解DNA的组成情况,也是核酸研究中最有用的物理参数之一。在温度为Tm时,DNA除了表现出增色效应外,同时还伴有比旋光值下降和黏度下降等现象。一般讲,RNA(核糖核酸)变性时的变性曲线不及DNA陡,Tm值也较低,tRNA有较多的双螺旋区,故有较高的Tm
