To construct the DNA vaccine against herpes simplex virus by using the fusion gene of herpes simplex virus type Ⅱ dominant epitope and hepatitis B core protein,the DNA of HSV-2 strain 333 was extracted.
提取HSV-2333株的DNA作为模板,用PCR方法扩增编码HSV-2糖蛋白D抗原优势表位的基因(gDt)和乙肝核心蛋白(HbcAg)的基因序列(HBc),再用重叠PCR方法将gDt插入到HBc编码Hb-cAg刺突部的第81和82位的氨基酸的基因之间,获得融合基因gDt-HBc,将gDt-HBc插入真核表达质粒pcDNA3。
