方法RT-PCR方法从小鼠淋巴细胞扩增出小鼠Foxp3基因,连入T Easy Vector进行测序,然后将序列正确的小鼠Foxp3基因用酶切的方法从T Easy vector切下,并将其连接到用相同酶切,含有硫氧还蛋白(含6个组氨酸“标签”)的pET 32a(+)原核表达载体中构建pET 32a(+)-MFoxp3表达载体。

The aims of this study were to investigate the pathogenesis from immunological regulation level through analysis of CD4~+CD25~+ T regulatory cells (Tregs) and forkhead transcription factor protein 3 (FOXP3) in patients with PBC by means of flow cytometry, RT-PCR and immune suppression analysis.
本研究以CD4~+CD25~+免疫调节性T细胞(T regulatory cell，Tregs)及其相关转录因子FOXP3为中心，通过对我国PBC患者人群Tregs细胞定量检测及FOXP3的基因和蛋白表达的分析、FOXP3逆转录病毒载体的构建、体外诱导免疫调节性T细胞及细胞增殖抑制实验等一系列体外细胞实验，在免疫调节水平上进一步探讨该病的发病机制。
