性质：利用特效可逆生物化学反应对大分子进行选择性分离纯化的一种与液相色谱法有关的技术。是利用生命现象中生物分子之间存在着非常特异的相互作用，如酶与底物、酶与抑制剂、酶与辅酶、激素与细胞受体、维生素与结合蛋白、基团与核酸、抗体与抗原等都相互具有亲和力，能形成可逆的配位化合物。利用可逆配位合物的结合与解离原理建立起来的一种简单、快速、回收率高的新的纯化技术。把能与待测大分子有特效配合作用的配位体（如酶或抗原等）接着在载体颗粒上组成固定相，流动相是溶于合适的缓冲溶液的试样，其中含有生物大分子（如酶抑制剂、抗体等）。当试样溶液通过柱后，再用同一缓冲溶液进行淋洗，除去各种杂质，再换用另一种不利于酶抑制剂相互作用的缓冲溶液，将抑制剂洗脱出。该法可以分离体内微量成分。用作纯化酶、酶的抑制剂、抗体、抗原、各种结合蛋白、激素和药物的受体、核酸、基团、多酶系统。并为研究生物体内各种分子相互作用及其机制提供有效的手段。
