性质:又称分光光度法,光吸收分析(1ight absorption analysis)。利用物质对光的选择性吸收进行定性和定量化学分析的总称。用白色光比较物质的色度的方法为比色分析,用单色光进行比较和测定的方法为吸光光度法。目前,在使用中并没有严格地区分。J.H.Lambert推导出表示光的吸收和物质层厚度的关系,即朗伯定律。其后,A. Beer推导出光的吸收与物质的浓度的关系,即朗伯-比尔定律,在此基础上,建立了今天的光吸收分析。一般每种物质仅吸收特定波长的光,利用这一性质,可作定性分析,或者利用朗伯-比尔定律,物质的吸光度同浓度成比例,根据物质吸光的程度、进行定量分析。目前广泛使用的比色管、比色计、光电光度计、光电分光光度计等,均属此类分析。分光光度法的选择性和灵敏度较比色法高。
