指持续应用激动剂时,受体的敏感性进行性下降,失去其正常功能,受体虽与激动剂结合,不发生受体蛋白构型的变化,离子通道不开放。脱敏感受体数量增加使具有正常去极化受体的总量减少,所产生的终板膜电位不足以引起肌纤维收缩的阈值,则神经肌肉兴奋传递就不再发生。其发生机制仍未十分清楚,可能为:①受体是一个大分子蛋白质,其许多部位可受小分子物质的影响;②受体处于肌膜的脂质中,两者间的相互作用;③某些物质影响受体蛋白构型。氟烷、多黏菌素、乙酰胆碱、新斯的明、利多卡因、氯丙嗪、巴比妥类药物可促使其发生。
